Characterization and discrimination of three Rhopalosiphum species (Homoptera: Aphididae) based on isozymes Zoologia

Seventeen clones of Rhopalosiphum padi (Linnaeus, 1758), 14 of R. maidis (Fitch, 1856), and two of R. insertum (Walker, 1849), representing a wide range of host plants and geographic distribution, were examined electrophoretically to determine intra and interspecific variation. Twenty-one enzyme systems were tested using starch-gel techniques. The electromorph variation within species was low, as expected for parthenogenetic organisms. Frequency of heterozygotes was also relatively low for most populations. Of eleven proteins analyzed in four Amazonian populations, the esterases showed the greatest variation, with five activity zones. EST1, EST2 and EST5 showed variation in each of the populations studied. EST1 and EST2 are each controlled by two, and EST5 by four, codomi-nant alleles. Changes in the expression of genes were observed during development in populations of Anopheles (Anopheles) intermedius and Anopheles (Anopheles) mattogrossensis. Enzymatic analysis in Anopheles nuneztovari Gabaldón (Diptera, Culicidae) BJB

Enzymatic analysis in Anopheles nuneztovari was made using four populations from the Brazilian Amazon and two from Colombia. The enzymes ME and XDH presented a monomorphic locus in all of the studied populations. EST and LAP presented a higher number of loci. In EST, genetic variation was observed in the five loci; LAP presented four loci, with allec variation in two loci. Dentre as diversas árvores frutíferas nativas dos cerrados, a cagaiteira (Eugenia dysenterica DC.) merece destaque pelo seu amplo potencial econômico. A fim de fornecer algumas informações relativas ao padrão espacial da variabilidade genética desta espécie, foram realizadas análises de autocorrelação espacial das freqüências alélicas em dez subpopulações locais da região sudeste do Estado de Goiás. Isolation and partial purification of beta-galactosidases from cotyledons of two cowpea cultivars Rev. Bras. Fisiol. Veg.

Three isoforms of beta-galactosidases were isolated and partially purified from the cotyledons of quiescent seeds of Vita 3 and Vita 5 cowpea [Vigna unguiculata (L.) Walp.] cultivars differing in water and salt stress tolerance. The purification procedure consisted of ammonium sulfate fractionation, acid precipitation, ion exchange chromatography through DEAE-sephadex and affinity chromatography through Lactosyl-sepharose columns. The migration rate of esterases and their substrate specificity for 4-methylumbelliferyl esters (acetate, propionate, and butyrate) and alpha- and beta-naphthyl esters were analyzed in Diatraea saccharalis by starch gel electrophoresis. Substrate preference of esterases was observed with Est-2 and Est-8 isozymes showing substrate specificity for 4-methylumbelliferyl esters and Est-4 isozyme showing specificity for 4-methylumbelliferyl butyrate and alpha-naphthyl butyrate. Uma população natural de Copaifera langsdorffii Desf., espécie arbórea comumente encontrada no Brasil, foi estudada por meio da eletroforese de isoenzimas, visando determinar a taxa de cruzamento e o sistema reprodutivo. A população amostrada localiza-se ao longo de um trecho do rio Capivari, em uma área de preservação permanente (floresta ciliar), entre os municípios de Lavras e Itumirim, sul do Estado de Minas Gerais, Brasil. Foram amostrados tecidos foliares de 20 indivíduos adultos da população e 400 indivíduos jovens (progênies) procedentes de sementes coletadas de 20 matrizes na população. Isoenzymatic variability of cassava accessions from different regions in Brazil Scientia Agricola

Cassava (Manihot esculenta Crantz) belongs to the Euphorbiaceae family, and is widely cultivated in Brazil. The objective of this study was to evaluate the isoenzymatic variability of 200 cassava accessions from the germplasm bank of Embrapa Amazonia Oriental. Seven groups were formed according to their origin: 1-Amazonas, 2-Amapa, 3-Bahia, 4-Para, 5-Rondonia, 6-Various, for accessions with a maximum of three individuals per place of origin, and 7 - Accessions of indefinite origin. Enzymatic variability in Aedes aegypti (Diptera: Culicidae) populations from Manaus-AM, Brazil Genet. Mol. Biol.

Eighteen enzymatic loci were analysed in Aedes aegypti populations from four neighbourhoods in the city of Manaus. The analyses showed that the Downtown population was the most polymorphic (p = 55.6%) with higher observed and expected mean heterozygosities (Ho = 0.152 ± 0.052; He = 0.174 ± 0.052, respectively). The least variability was detected in the Coroado and Cidade Nova populations, both with polymorphism of 44.4%. The latter population presented the least observed heterozygosity (Ho = 0.109 ± 0.037). Isozyme variation in four species of the Simulium perflavum species group (Diptera: Simuliidae) from the Brazilian Amazon Genet. Mol. Biol.

Electrophoretic studies of isozymes were done with four closely related species of the Simulium perflavum species group (Diptera: Simuliidade) in the Brazilian Amazon, using last-instar larvae collected in the field. Ten enzymes were studied, which yielded 11 loci. Diagnostic loci were not found between Simulium maroniense cytotype D and Simulium rorotaense. Simulium maroniense and S. rorotaense differed from Simulium trombetense by two diagnostic loci (Me and Xdh), and Simulium perflavum differed from the other three species by four diagnostic loci (Me, Xdh, Mdh, and Got). Isoenzymatic variability in tropical maize populations under reciprocal recurrent selection Scientia Agricola

Maize (Zea mays L.) is one of the crops in which the genetic variability has been extensively studied at isoenzymatic loci. The genetic variability of the maize populations BR-105 and BR-106, and the synthetics IG-3 and IG-4, obtained after one cycle of a high-intensity reciprocal recurrent selection (RRS), was investigated at seven isoenzymatic loci. A total of twenty alleles were identified, and most of the private alleles were found in the BR-106 population. Mating system in a neotropical tree species, Senna multijuga (Fabaceae) Genet. Mol. Biol.

Senna multijuga is a pioneer tropical tree species that occurs mainly in the Brazilian Atlantic forest. We investigated the mating system of two populations of S. multijuga, one located in a reserve area (RD1) and the other (RD2) about 15 km away. The mating system parameters were estimated using the mixed mating model (software MLTR). The two populations had significantly different outcrossing rates, with population RD2 having a high rate (t m = 0.838) and population RD1, a lower rate (t m = 0.540). Polyacrylamide gel electrophoresis, RAPD techniques and isozymes of esterase, phosphoglucomutase and soluble proteins were analyzed in tubers, and isocitrate dehydrogenase, aspartate transaminase, phosphoglucomutase and peroxidase, in leaves. Esterase isozymes were used to detected substrate-preference polymorphism in twenty cultivars of Manihot esculenta, and to show cultivar-specific variation of this species. A relatively complex extraction solution of proteins from leaves was needed to show a larger number of esterase isozymes. Similarity between cultivars from six groups ranged from 51 to 96%. The cultivars identified by the same name seemed to be biochemically different regarding esterase isozymes. The aim of this study was to delimit two Iodophanus species: I. carneus and I. testaceus , based on morphological characteristics and electrophoretic patterns of their intracellular isozymes. Twenty monosporic strains were used, including five belonging to I. granulipolaris as a control. Fourteen isozyme systems were tested, and the five having the best resolution selected: aspartate aminotransferase, esterases, alkaline phosphatase, glutamate dehydrogenase, and superoxide dismutase. These analyses confirmed the similarity between I. carneus and I. testaceus , since they both produced the same band patterns, which were in turn different from the band pattern of I. granulipolaris.
